Some writers make detailed notes about their characters, even going so far as to know what the character's favorite color, snack and movie is.
Others start with a notion or vague concept of a character and discover who the person is as they get deeper into the screenplay.
And others work somewhere inbetween.
Which way is best? Depends on the person. Depends on how much you, as the writer, feel you need to know about someone before you begin putting words in his mouth and actions specific to him.
I personally like to find out who my characters are as they move through the script. This works for me because by the time I've finished the first draft I've learned things about her that I didn't know at the start.
